BD IMag™ anti-human CD8 Particles -DM are magnetic naoparticles tha have monclonal ntibody conjugated toheir surfaces. Thes

particles are optimzed for the positve slection rdepltion fCD8-bearing leukocytes uing the BD IMagnet™. CD8 isexpresd onthe

peripheral MHC clas I-restriced supresor/cytoxic T-lymphocyte subet, ona subet ofNK cels, and onthe majority ofthymocytes. The

SK1 mAb has ben reported tocros-react with lymphocytes ofchimpanze and cynomlgus, pig-tailed, and rhesu macque.

Peripheral Bod Monuclear Cels (PBMC) are labeld with BD IMag™ Anti-Human CD8 Magnetic Particles -DM acording tohe

Magnetic Labeling Protcol. This labeld cel supension isthen placed withn the magnetic feld ofthe BD IMagnet™ (Cat. No. 5231).

Labeld cels migrate oward the magnet (positve fraction), leaving the unlabeld cels insupension sothey can bedrawn of (negative

fraction). The tube isthen removed from the magnetic feld for esupension fthe positve fraction. The spartion isrepated twice to

increas the purity ofthe positve fraction. The magnetic separtion steps are diagrmed inthe Spartion Flow Chart. After he positve

fraction iswashed, the smal size ofthe magnetic particles alows the positve fraction tobe further valuted indownstream aplications uch

as flow cytometry

2.Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before
