Background Information CD45 is a receptor-tyrosine phosphatase required for antigen-induced signaling in T-cells and B-cells; its substrates are unknown. Interestingly, it associates with several Src-family tyrosine kinases, which are inhibited by tyrosine phosphorylation in their c-termini, suggesting a mechanism for signaling by CD45.

Product Information
Format Purified
Control
  • RAW264 Cell Lysate
Presentation Purified mouse monoclonal IgG2b in phosphate buffered saline containing 10 mM sodium azide and 1 mg/mL bovine serum albumin.
We recommend that each laboratory determine an optimum working titre for use in its particular application.

Applications
Application Anti-CD45RA Antibody, Clone MEM 56 detects level of CD45RA & has been published & validated for use in FC, IF, IH(P), IP & WB.
Key Applications
  • Flow Cytometry
  • Western Blotting
  • Immunoprecipitation
  • Immunohistochemistry (Paraffin)
  • Immunofluorescence
Application Notes Immunofluorescence: Identification of CD4+ suppressor/inducer cells, by indirect immunofluorescence staining.

Immunoprecipitation: A previous lot of this antibody detected 220 kDa molecular weight component of the leucocyte common antigen complex in immunoprecipitation.

Flow Cytometry: Purification or depletion of CD4+ suppressor/inducer cells by cell sorting or the panning techniques was performed on a previous lot.

Immunohistochemistry(paraffin): Representative testing from a previous lot.
Identification of CD45R+ lymphocytes in cryostat sections of frozen tissue and in formalin fixed wax embedded tissue.

Optimal Staining of CD45RA Monoclonal Antibody: Tonsil

Concentration Please refer to the Certificate of Analysis for the lot-specific concentration.
Host Mouse
Specificity This antibody recognizes the 220 kDa molecular weight component of the leucocyte common antigen complex expressed on some CD4+ T lymphocytes, B lymphocytes and CD8+ lymphocytes. The CD45R+/CD4+ lymphocytes functionally exert suppressor/inducer activity in in vitro assay systems.
Antigen distribution:
B-cells (lg+) > 95%
T-cells (E+) 70 + 10%
Peripheral blood lymphocytes 68 + 8%
T-cells (CD4+) 50 + 10%
Monocytes 50 + 20%
Thymocytes < 5%
Granulocytes < 1%
Isotype IgG2b
Species Reactivity Human Rat Mouse
Species Reactivity Note Mouse. Expected to react with human and rat based on sequence homology. .

Entrez Gene Number
Entrez Gene Summary The protein encoded by this gene is a member of the protein tyrosine phosphatase (PTP) family. PTPs are known to be signaling molecules that regulate a variety of cellular processes including cell growth, differentiation, mitotic cycle, and oncogenic transformation. This PTP contains an extracellular domain, a single transmembrane segment and two tandem intracytoplasmic catalytic domains, and thus belongs to receptor type PTP. This gene is specifically expressed in hematopoietic cells. This PTP has been shown to be an essential regulator of T- and B-cell antigen receptor signaling. It functions through either direct interaction with components of the antigen receptor complexes, or by activating various Src family kinases required for the antigen receptor signaling. This PTP also suppresses JAK kinases, and thus functions as a regulator of cytokine receptor signaling. Four alternatively spliced transcripts variants of this gene, which encode distinct isoforms, have been reported.

UniProt Number
UniProt Summary FUNCTION: SwissProt: P08575 # Required for T-cell activation through the antigen receptor. The first PTPase domain has enzymatic activity, while the second one seems to affect the substrate specificity of the first one. Upon T-cell activation, recruits an dephosphorylates SKAP1 and FYN.
SIZE: 1304 amino acids; 147254 Da
SUBUNIT: Binds GANAB and PRKCSH (By similarity). Interacts with SKAP1.
SUBCELLULAR LOCATION: Membrane; Single-pass type I membrane protein.
DOMAIN: SwissProt: P08575 The first PTPase domain interacts with SKAP1.
PTM: Heavily N- and O-glycosylated.
DISEASE: SwissProt: P08575 # Defects in PTPRC are a cause of autosomal recessive severe combined immunodeficiency T-cell-negative/B-cell- positive/NK cell-positive (T(-)/B(+)/NK(+) SCID) [MIM:608971]. SCID refers to a genetically and clinically group of rare congenital disorders characterized by impairment of both humoral and cell-mediated immunity, leukopenia, and low or absent antibody levels. Patients with SCID present in infancy with recurrent, persistent infections by opportunistic organisms, including Candida albicans, Pneumocystis carinii, and cytomegalovirus, among many others. The common characteristic of all types of SCID is absence of T-cell-mediated cellular immunity due to a defect in T- cell development. & Genetic variations in PTPRC are involved in multiple sclerosis susceptibility (MS) [MIM:126200]. MS is a neurodegenerative disorder characterized by the gradual accumulation of focal plaques of demyelination particularly in the periventricular areas of the brain. Peripheral nerves are not affected. Onset usually in third or fourth decade with intermittent progression over an extended period. The cause is still uncertain.
SIMILARITY: Belongs to the protein-tyrosine phosphatase family. Receptor class 1/6 subfamily. & Contains 2 fibronectin type-III domains. & Contains 2 tyrosine-protein phosphatase domains.
