PI3 Kinase (Phosphatidylinositol 3-Kinase) is responsible for phosphorylation of the 3 position of the inositol ring of PI(4,5)P2, to generate PI(3,4,5)P3, a potent second messenger required for survival signaling, and insulin action. PI3 Kinase is a heterodimeric complex composed of an 85 kDa regulatory subunit and a 110 kDa catalytic subunit. Tyrosine phosphorylation of growth factor receptors creates docking sites for binding of p85 (through its SH2 domains) on the receptors; p85 brings with it p110, which is then proximal to its phospho-lipid substrate on the membrane. PI3 Kinase is also activated by Ras, and by the β:γ subunits of heterotrimeric G-proteins. PI3 Kinase is inhibitable by wortmannin, a useful tool for the study of the PI3 Kinase signaling pathway.

Phosphoinositide 3-kinases (PI3Ks) phosphorylate the 3-prime OH position of the inositol ring of inositol lipids. See MIM 602838. The class I PI3Ks display a broad phosphoinositide lipid substrate specificity and include p110-alpha (MIM 171834), p110-beta (MIM 602925), and p110-gamma (MIM 601232). p110-alpha and p110-beta interact with SH2/SH3-domain-containing p85 adaptor proteins (see MIM 171833) and with GTP-bound Ras.[supplied by OMIM]
SIZE: 1044 amino acids; 119479 Da
SUBUNIT: Heterodimer of a p110 (catalytic) and a p85 (regulatory) subunit. Interacts with ERAS (By similarity).
TISSUE SPECIFICITY: Expressed predominantly in leukocytes.
PTM: Autophosphorylation on Ser-1039 results in the almost complete inactivation of the lipid kinase activity.
SIMILARITY: SwissProt: O00329 ## Belongs to the PI3/PI4-kinase family. & Contains 1 PI3K/PI4K domain.
Product Usage Statements
Quality Assurance
Routinely evaluated by Western Blot on Jurkat cell lysate.
Western Blot Analysis:
A 1:500 dilution of this lot detected endogenous PI3 Kinase, p110δ in lysate from Jurkat cells.
Usage Statement
Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage Conditions
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations:
Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20ºC. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20ºC may cause glycerol-containing solutions to become frozen during storage.
