Background Information Cyclins are, as their name suggests, expressed at specific times during the cell cycle, and play a crucial role in the regulation of the cell cycle. Cyclins associate with cyclin-dependent kinases (cdks), and the association is absolutely required for cdk activity. At the conclusion of the relevant phase of the cell cycle, cyclins are rapidly ubiquitinated and degraded, thus returning cdks to their inactive state. Cyclin A associates with cdk2, and together with cyclin E is involved in the G1/S transition. Cyclin A/cdk2 remains active during S-phase. Cyclin B associates with cdk1/cdc2 to regulate the G2/M boundary. The D-cyclins bind cdk4 and cdk6, and result in Rb phosphorylation. The cyclin D1 gene was found in chronic B cell lymphocytic leukemia, and was designated Bcl-1 in a previously used nomenclature.

Entrez Gene Number
Entrez Gene Summary The protein encoded by this gene belongs to the highly conserved cyclin family, whose members are characterized by a dramatic periodicity in protein abundance throughout the cell cycle. Cyclins function as regulators of CDK kinases. Different cyclins exhibit distinct expression and degradation patterns which contribute to the temporal coordination of each mitotic event. This cyclin forms a complex with and functions as a regulatory subunit of CDK4 or CDK6, whose activity is required for cell cycle G1/S transition. This protein has been shown to interact with tumor suppressor protein Rb and the expression of this gene is regulated positively by Rb. Mutations, amplification and overexpression of this gene, which alters cell cycle progression, are observed frequently in a variety of tumors and may contribute to tumorigenesis. [provided by RefSeq]

UniProt Number
UniProt Summary Function: Essential for the control of the cell cycle at the G1/S (start) transition.
Subunit structure: Interacts with the CDK4 and CDK6 protein kinases to form a serine/threonine kinase holoenzyme complex. The cyclin subunit imparts substrate specificity to the complex. Ref.10
Involvement in disease: A chromosomal aberration involving CCND1 may be a cause of B-lymphocytic malignancy, particularly mantle-cell lymphoma (MCL). Translocation t(11;14)(q13;q32) with immunoglobulin gene regions. Activation of CCND1 may be oncogenic by directly altering progression through the cell cycle.
A chromosomal aberration involving CCND1 may be a cause of parathyroid adenomas [MIM:168461]. Translocation t(11;11)(q13;p15) with the parathyroid hormone (PTH) enhancer.
A chromosomal aberration involving CCND1 may be a cause of multiple myeloma [MIM:254500]. Translocation t(11;14)(q13;q32) with the IgH locus.
Sequence similarities: Belongs to the cyclin family. Cyclin D subfamily

Product Usage Statements
Quality Assurance Routinely evaluated by western blot on MCF-7 cell lysate.

Western Blot Analysis:
A 1:1,000 dilution of this lot was used to detect Cyclin D1 on MCF-7 cell lysate.
Usage Statement
  • Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.

Storage and Shipping Information
Storage Conditions Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
