Jun is a component of the transcription factor AP-1, and can constitute AP-1 activity either as a homodimer or as a heterodimer with Fos. By contrast with Fos, Jun is expressed constitutively, and can be activated by phosphorylation of serines 63 and/or 73. These sites are targets for the SAPK1/JNK family of protein kinases, and their phosphorylation states can be monitored using antibodies specific for the phospho-epitopes. In addition, Jun is phosphorylated by GSK3 on sites which inhibit Jun activity. Since GSK3 is inactivated by Akt, the Akt pathway results in Jun activation.

Application Notes
Immunohistochemistry:
1:250 dilution of a previous lot was used in immunohistochemistry (antigen retrieval required).
Immunoprecipitation:
1:40 dilution of a previous lot was used in immunoprecipitation.
Immunocytochemistry:
1:250 dilution of a previous lot was used in immunocytochemistry.
Biological Information
Immunogen
Synthetic conjugated peptide directed against the N-terminus of human c-Jun.
This gene is the putative transforming gene of avian sarcoma virus 17. It encodes a protein which is highly similar to the viral protein, and which interacts directly with specific target DNA sequences to regulate gene expression. This gene is intronless and is mapped to 1p32-p31, a chromosomal region involved in both translocations and deletions in human malignancies.
FUNCTION:SwissProt: P05412 # Transcription factor that recognizes and binds to the enhancer heptamer motif 5'-TGA[CG]TCA-3'. SIZE:331 amino acids; 35676 Da SUBUNIT:Interacts with FOS to form a dimer. Interacts with SMAD3/SMAD4 heterodimers. Interacts with MYBBP1A, SPIB and TCF20. Interacts with COPS5; indirectly leading to its phosphorylation. Interacts with DSIPI; this interaction inhibits the binding of active AP1 to its target DNA. SUBCELLULAR LOCATION:Nucleus. PTM:Phosphorylation enhances the transcriptional activity. Phosphorylated by PRKDC. SIMILARITY:SwissProt: P05412 ## Belongs to the bZIP family. Jun subfamily. & Contains 1 bZIP domain.
Product Usage Statements
Quality Assurance
Routinely evaluated by Western Blot on NIH/3T3 whole cell lysate.
Western Blot Analysis:
A 1:1,000-1:2,000 dilution of this lot was used to detect c-Jun on NIH/3T3 whole cell lysate.
Usage Statement
Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage Conditions
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
