Interleukin-13 (IL-13) is a cytokine that is secreted by helper T cells in reaction to T cell antigen receptor signaling. IL-13 is also secreted by mast cells and basophils when cross-linking with immunoglobulin E. IL-13 seems to play a role as an effector in ulcerative colitis that weakens epithelial barrier function by influencing epithelial apoptosis, tight junctions and restitution velocity. IL-13 has been shown to be involved in the pathogenesis of allergic diseases such as atopic dermatitis. IL-13 seems to also have a role to play in alcohol mediated activation of pro-fibrotic pathways. IL-13 is thought to be an important cytokine during the expansion of asthma and chronic obstructive pulmonary disease and may be critical clinical therapy targets.

This gene encodes an immunoregulatory cytokine produced primarily by activated Th2 cells. This cytokine is involved in several stages of B-cell maturation and differentiation. It up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. This cytokine down-regulates macrophage activity, thereby inhibits the production of pro-inflammatory cytokines and chemokines. This cytokine is found to be critical to the pathogenesis of allergen-induced asthma but operates through mechanisms independent of IgE and eosinophils. This gene, IL3, IL5, IL4, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL4. [provided by RefSeq].
FUNCTION: Cytokine. Inhibits inflammatory cytokine production. Synergizes with IL2 in regulating interferon-gamma synthesis. May be critical in regulating inflammatory and immune responses.
SUBCELLULAR LOCATION: Secreted.
INVOLVEMENT IN DISEASE: Defects in IL13 may be a cause of susceptibility to allergic rhinitis (ALRH) [MIM:607154]. Allergic rhinitis is a common disease of complex inheritance and is characterized by mucosal inflammation caused by allergen exposure.
SEQUENCE SIMILARITIES: Belongs to the IL-4/IL-13 family.
CAUTION: It is uncertain whether Met-1 or Met-15 is the initiator.
SEQUENCE CAUTION: The sequence AAA36107.1 differs from that shown. Reason: Erroneous initiation. Translation N-terminally extended.
