Background Information
The Ca2+/Calmodulin-Dependent Kinase family includes CaM Kinases I through IV, Phosphorylase Kinase, and MLCK. The CaM Kinases are inactive in the absence of stimuli; binding to Ca2+/Calmodulin induces conformational changes and activation. CaM Kinases have broad substrate specificity, with many cellular functions.

Immunocytochemistry
Application Notes
Immunohistochemistry Analysis: A 1:100-250 dilution from a previous lot detected Calcium/Calmodulin Kinase I in human kidney tissue.
Immunocytochemistry Analysis: A 1:100-250 dilution from a previous lot was used in IC.
Biological Information
Immunogen
Specific peptide corresponding to residues on the N-terminus of human Calcium/Calmodulin Kinase I.
Epitope
N-terminus
Clone
EPR2217Y
Host
Rabbit
Specificity
This antibody recognizes Calcium/Calmodulin Kinase I.
Calcium/calmodulin-dependent protein kinase I is expressed in many tissues and is a component of a calmodulin-dependent protein kinase cascade. Calcium/calmodulin directly activate calcium/calmodulin-dependent protein kinase I by binding to the enzyme and indirectly promotes the phosphorylation and synergistic activation of the enzyme by calcium/calmodulin-dependent protein kinase I kinase.
FUNCTION:Calcium/calmodulin-dependent protein kinase belonging to a proposed calcium-triggered signaling cascade involved in a number of cellular processes. Isoform 1, isoform 2 and isoform 3 phosphorylate CAMK1 and CAMK4. Isoform 3 phosphorylates CAMK1D. Isoform 4, isoform 5 and isoform 6 lacking part of the calmodulin-binding domain are inactive. Seems to be involved in hippocampal activation of CREB1 By similarity. CATALYTIC ACTIVITY:ATP + a protein = ADP + a phosphoprotein. ENZYME REGULATION:Activated by Ca2+/calmodulin. Binding of calmodulin may releave intrasteric autoinhibition. Autophosphorylation does not alter activity or regulation by Ca2+/calmodulin. In part, activity is independent on Ca2+/calmodulin By similarity. SUBUNIT STRUCTURE:Interacts with calmodulin By similarity. SUBCELLULAR LOCATION:Cytoplasm By similarity. TISSUE SPECFICITY:Ubiquitously expressed with higher levels in the brain. Intermediate levels are detected in spleen, prostate, thyroid and leukocytes. The lowest level is in lung. DOMAIN:The autoinhibitory domain overlaps with the calmodulin binding region and may be involved in intrasteric autoinhibition.
The RP domain (arginine/proline-rich) is involved in the recognition of CAMKI and CAMK4 as substrates By similarity. PTM:Autophosphorylated By similarity. SEQUENCE SIMILARITIES:Belongs to the protein kinase superfamily. Ser/Thr protein kinase family.
Contains 1 protein kinase domain. SEQUENCE CAUTION:The sequence CAD38990.1 differs from that shown. Reason: Miscellaneous discrepancy. Intron retention.

Quality Assurance
Evaluated by Western Blot on Human brain, mouse brain, rat brain, and SHSY5Y lysates.
Western Blot Analysis: A 1:5,000-10,000 dilution of this antibody was used to detect Calcium/Calmodulin Kinase I in Human brain, mouse brain, rat brain, and SHSY5Y lysates.
Usage Statement
Unless otherwise stated in our catalog or other company documentation accompanying the product(s), our products are intended for research use only and are not to be used for any other purpose, which includes but is not limited to, unauthorized commercial uses, in vitro diagnostic uses, ex vivo or in vivo therapeutic uses or any type of consumption or application to humans or animals.
Storage and Shipping Information
Storage Conditions
Stable for 1 year at -20ºC from date of receipt.
Handling Recommendations: Upon first thaw, and prior to removing the cap, centrifuge the vial and gently mix the solution. Aliquot into microcentrifuge tubes and store at -20°C. Avoid repeated freeze/thaw cycles, which may damage IgG and affect product performance. Note: Variability in freezer temperatures below -20°C may cause glycerol containing solutions to become frozen during storage.
