Myotonic dystrophy (DM1) is a neuromuscular disorder associated with repeated CTG triplets in the 3'-untranslated region of the DMPK gene. It is hypothesized that the CUG repeats in the mRNA form hairpins that sequester nuclear factors required for proper muscle development and maintenance. MBNL (muscleblind-like protein 1) is an RNA splicing factor that binds to the CUG repeats and is enriched at these mutant RNA foci. MBNL-deficient mice are often used as a model for myotonic muscular dystrophy and siRNA downregulation of MBNL in normal myoblasts results in abnormal insulin receptor splicing.
