Recombinant mouse EPHA1 (amino acids 569–end) was expressed by baculovirus in Sf9 insect cells using an N-terminal GST tag. EPHA1 is a member of the Eph family of receptor tyrosine kinases that have been implicated in mediating developmental events, particularly in the nervous system. Receptors in the Eph subfamily typically have a single kinase domain and an extracellular region containing a Cys-rich domain and 2 fibronectin type III repeats. EPHA1 seems to be a marker of the differentiated normal epidermis, and its downregulation in nonmelanoma skin cancer may contribute to carcinogenesis of these very frequent human tumors. EPHA1 represents a new potential prognostic marker and therapeutic target in nonmelanoma skin cancer.
