Figure. DharmaFECT Duo achieves optimal co-transfection results - effectiv plasmid delivery (black bars) and target mRNA knockdown (blue bars) when compared to other lipid reagents. The psiCHECK 2 Vector (Promega; 100 ng/well) and siRNA targeting renilla luciferase (100 nM) were complexed with a range of transfection reagent volumes (0.05 - 0.6 μL/well) to determine optimal co-transfection conditions (shown). Firefl y Luciferase expression was assessed at 48h using the Dual-Glo™ Luciferase Assay System (Promega). Cell viability was assessed using the alamarBlue® Assay (Biosource International). siRNA-induced Renilla luciferase knockdown (blue bars) was assessed by branched DNA assay (Panomics).
In side-by-side co-transfection experiments with traditional reagents developed for plasmid delivery, only Thermo Scientific DharmaFECT Duo Transfection Reagent resulted in efficient cell delivery of both siRNA (assayed by target gene knockdown) or microRNA reagents, along with plasmid (assayed by level of reporter expression). DharmaFECT Duo is an excellent choice for experiments requiring co-transfection, such as RNAi rescue or utilization of reporter systems.
