DharmaFECT 1,2,3和4系列产品是专门为不同细胞系转染siRNA而优化的高效转染试剂。众所周知,单一的转染试剂不可能满足上百种细胞系对siRNA的转染要求;因此,Dharmacon通过对上百种细胞系/株逐个筛选和优化,开发出了最适合特定细胞系/株的转染试剂,满足更大范围的细胞系/株对siRNA的高效转染需求(见下图)。


Figure.DharmaFECT siRNA transfection reagents are very effective in delivering siRNA with minimal effect on cell viability. DharmaFECT formulations 1-4(DF1, DF2,DF3, and DF4) were used to transfect 100 nM siRNA targeting GAPDH in a variety of cell lines.At 72 hours, GAPDH expression (bars) was determined by branched DNA assay (Panomics Quantigene® Reagent System) and cell viability (data points) was determined by alamarBlue™ (Biosource International).

 

在不同的转染条件下,DharmaFECT转染试剂与其他品牌转染试剂相比较,更加容易找到试验的优化条件,并维持细胞高存活率的基础上达到高效的基因沉默效果。下图表示的试验为利用HepG2细胞在三种不同细胞浓度以及6中不同转染试剂浓度下转染100nM终浓度GAPDH的siRNA后,测定GAPDH基因mRNA水平和细胞存活率。


Figure . DharmaFECT (A) is effective across a broader range of experimental conditions when compared to a lipid from a leading manufacturer (B). Several cell densities and lipid volumes were investigated to determine optimal transfection conditions, shown by the shaded blue boxes. Three cell densities of HepG2 cells were transfected with GAPD siRNA (100nM) using a range of volumes of DharmaFECT 4 and a lipid from a leading manufacturer. GAPD mRNA levels (bars) were assessed by branched DNA assay (Panomics uantigene® Reagent System) and cell viability (data points) was determined by alamar- Blue™ (Biosource International)."
