Background:CXCL12/SDF-1 (PBSF)
Stromal cell-derived factor (SDF)-1 alpha and SDF-1 beta are the first cytokines initially identified using the signal sequence trap cloning strategy from a mouse bone-marrow stromal cell line. They are encoded by a single gene, PBSF, and arise by alternative splicing. The two proteins are identical except for the four amino acid residues that are present in the carboxy-terminus of SDF-1 beta and absent from SDF-1 alpha. SDF-1/PBSF is highly conserved between species. Human mature SDF-1 shares approximately 99% amino acid sequence identity with the mouse protein.

The Quantikine Mouse SDF-1 alpha Immunoassay is a 4.5 hour solid-phase ELISA designed to measure mouse SDF-1 alpha in cell culture supernates and mouse serum. It contains E. coli-expressed recombinant mouse SDF-1 alpha and antibodies raised against the recombinant factor. It has been shown to accurately quantitate recombinant mouse SDF-1 alpha. Results obtained using natural SDF-1 alpha showed linear curves that were parallel to the standard curves obtained using the Quantikine kit standards. These results indicate that this kit can be used to determine relative mass values of natural mouse SDF-1 alpha.
