Easy-to-Run Sandwich ELISA A monoclonal capture antibody specific for The Caspase-3 (regardless of phosphorylation state) has been coated onto the wells of the 96-well plate provided. Samples, including a standard of known The Caspase-3 (Active) content, controls, and unknowns, are pipetted into these wells and then a rabbit detection antibody specific for The Caspase-3 (Active) is added to the wells. During the first incubation, the The Caspase-3 antigen binds to the immobilized capture antibody and the The Caspase-3 rabbit antibody serves as a detection antibody by binding to the immobilized The Caspase-3 (Active) protein. After washing, a horseradish peroxidase labeled anti-rabbit IgG is added. This binds to the detection antibody to complete the four member sandwich. After a third incubation and washing to remove all the unbound enzyme, a substrate solution (TMB) is added, which is acted upon by the bound enzyme to produce color. The intensity of this colored product is directly proportional to the concentration of The Caspase-3 (Active) present in the original specimen and the optical density can be read on a standard microplate reader. The total assay incubation time is only 4 hours.
