HeLa cells were co-transfected with a constant concentration of siRNA (100 nM) comprised of increasing ratios of Cyclophilin B siRNA:RISC-Free™ (or Non-Targeting) siRNA. For comparison, a parallel set of samples included cells transfected with cyclophilin B siRNA alone. After 24 hours, cyclophilin B mRNA levels were measured using a branched DNA assay (Genospectra).When unmodified, nontargeting siRNA is included in the transfection mix, the level of targeted gene silencing is reduced in a dose-dependent fashion suggesting competition for the silencing machinery. In contrast, when RISC-Free™ siRNA is a part of the transfection mix, no effect on the silencing efficiency (no competition) is observed.

The Thermo Scientific siGLO Red Transfection Indicator is a fluorescent oligonucleotide duplex that localizes to the nucleus, thus permitting unambiguous visual assessment of uptake into mammalian cells. It is ideal for use in experiments to determine optimal siRNA transfection conditions and for monitoring relative delivery efficiency during silencing experiments. Transfection Indicators are intended as a qualitative indicator of delivery and should not be used for quantitative determination of siRNA uptake.
siGLO Red Transfection Indicator (DY-547) may show some punctate staining, but has a stronger signal at lower concentration than siGLO Green. It is suitable for transfection, electroporation or nucleofection.
