Figure . Silencing efficiency of siCONTROL™ Cyclophilin B siRNA in the presence of increasing amounts of siGLO™ RISC-Free™ siRNA. HeLa cells were co-transfected with a constant concentration of siRNA (100 nM) comprised of increasing ratios of Cyclophilin B siRNA:RISC-Free™ (or Non-Targeting) siRNA. For comparison, a parallel set of samples included cells transfected with cyclophilin B siRNA alone. After 24 hours, cyclophilin B mRNA levels were measured using a branched DNA assay (Genospectra).When unmodified, nontargeting siRNA is included in the transfection mix, the level of targeted gene silencing is reduced in a dose-dependent fashion suggesting competition for the silencing machinery. In contrast, when RISC-Free™ siRNA is a part of the transfection mix, no effect on the silencing efficiency (no competition) is observed.
Thermo Scientific siGLO RISC-Free Control siRNA is chemically modified to impair processing and uptake by RISC, allowing researchers to isolate cellular effects related to siRNA transfection. As transfection reagents typically rely on charged interaction with the nucleic acid payload, transfection controls using siGLO RISC-Free are recommended over the use of transfection reagent alone.
