SignalStain® Apoptosis (Cleaved Caspase-3) IHC Detection Kit 能够检测内源的Asp175位断裂的活化的caspase-3大片段(17/19 kDa)。该抗体不能识别全长caspase-3或其他的断裂caspases。仅推荐该试剂盒用于免疫组化分析。Cleaved Caspase-3 (Asp175) (D3E9) Rabbit mAb 兔单抗是采用合成的人caspase-3蛋白Asp175邻近氨基酸残基序列相应的肽段免疫动物制备的。Rabbit (DA1E) mAb IgG XP® SignalStain® Isotype Control不能直接与任何已知的抗原反应。它的功能是作为rabbit IgG单抗的同型对照。SignalStain® Apoptosis (Cleaved Caspase-3) IHC Detection Kit 能够检测福尔马林固定石蜡包埋的人和小鼠组织中活化的caspase-3. 多聚的HRP-偶联的SignalStain® Boost IHC Detection Reagent 配合 SignalStain® DAB Diluent 和 Chromogen Concentrate 检测 Cleaved Caspase-3 (Asp175) (D3E9) Rabbit mAb 兔单抗。同时，还要加入相应浓度的rabbit monoclonal IgG control 来确认染色的特异性。这种组合试剂提供了一个灵敏且特异的方式检测组织样品中的凋亡过程。Caspase-3 (CPP-32, Apoptain, Yama, SCA-1)是一个凋亡的关键执行者，因为它对许多关键的蛋白起着部分或全部的裂解作用，比如核酶多聚(ADP-ribose)聚合酶(PARP)（1）。caspase-3的活化需要经过其未激活的的酶原转变成活化的 p17 和 p12 片段的蛋白水解过程。caspase-3的剪切需要天门冬氨酸位于p1位点（2）。

Specificity / Sensitivity

SignalStain^® Apoptosis (Cleaved Caspase-3) IHC Detection Kit detects endogenous levels of the activated caspase-3 large fragment (17/19 kDa) resulting from cleavage adjacent to Asp175. This antibody does not recognize full-length caspase-3 or other cleaved caspases. This kit was developed for and is recommended for immunohistochemistry only.

Source / Purification

Cleaved Caspase-3 (Asp175) (D3E9) Rabbit mAb is produced by immunizing animals with a synthetic peptide corresponding to residues surrounding Asp175 of human caspase-3 protein. Rabbit (DA1E) mAb IgG XP^®SignalStain^® Isotype Control is not directed against any known antigen. It functions as an isotype control for rabbit IgG monoclonal antibodies.

Description

SignalStain^® Apoptosis (Cleaved Caspase-3) IHC Detection Kit allows the detection of activated caspase-3 in formalin-fixed paraffin-embedded human and mouse tissue samples. Cleaved Caspase-3 (Asp175) (D3E9) Rabbit mAb is detected by the polymer based, HRP-conjugated SignalStain^® Boost IHC Detection Reagent in combination with SignalStain^® DAB Diluent and Chromogen Concentrate. Also included is a concentration-matched rabbit monoclonal IgG control to verify the specificity of staining.This combination of reagents provides a sensitive and specific means of detecting apoptotic events in tissue samples.

Background

Caspase-3 (CPP-32, Apoptain, Yama, SCA-1) is a critical executioner of apoptosis, as it is either partially or totally responsible for the proteolytic cleavage of many key proteins, such as the nuclear enzyme poly (ADP-ribose) polymerase (PARP) (1). Activation of caspase-3 requires proteolytic processing of its inactive zymogen into activated p17 and p12 fragments. Cleavage of caspase-3 requires the aspartic acid residue at the P1 position (2).
