Reconstitute with 2 ml distilled water.
Ethanol or methanol may be used instead of water.
Intended Use
DAPI is an indole dye that binds strongly and selectively to the minor groove of adenine-thymine regions of DNA. When bound to double-stranded DNA it absorbs light at 340nm (ultraviolet light) and emits at 488nm (fluoresces blue). Bound DAPI has a fluorescence intensity approximately 20 fold higher than that of unbound DAPI. The fluorescence is also directly proportional to the amount of DNA present.
DAPI also binds to RNA via a slightly different mechanism, with an emission maximum at ~500nm. The fluorescence is less intense (~20%) than that of DAPI-bound double stranded DNA.
DAPI is cell-permeable and is commonly used to detect nuclei and organellar DNA in immunofluorescence, histochemical andin situhybridisation procedures. It may also be used to detect mycoplasmas and other microbial contamination, assessing apoptosis, assaying DNA in solution and measuring nuclear content and sorting chromosomes in flow cytometry. DAPI has also been used to replace ethidium bromide to stain DNA in agarose gels and may be used as a vital dye to stain mature pollen grains.
