CD27 is a member of the TNF-receptor family. The expression of CD27 is restricted to cells of the lymphoid lineage and appears to be related to functional differentiation programs of T- and B-cells. Constitutively expressed on T-lymphocytes, CD27 is a costimulatory molecule for a regulatory subset. Induced on B lymphocytes after antigenic challenge, it is a marker of memory cells. Following antigen activation in germinal centers, B-cells develop into memory B-cells or plasma cells. CD27 interacts with its ligand CD70 to control the entry of B cells into their pathway. Structurally CD27 being a member of the TNF receptor superfamily is related to Fas, CD40 and RANK. These receptors are characterized by an extra cellular domain comprised of two to six repeats of cysteine rich motifs and the existence of a number of structurally related decoy receptors.
A soluble isoform of CD27, sCD27 has been described which is found in body fluids and can be used to monitor local and systemic immune activation. In addition elevated serum concentration of sCD27 are found in patients with B-cell malignancies and levels of sCD27 strongly correlate with tumor load and neoplastic B-cell growth. It was furthermore shown to be a marker increased in patients suffering from a variety of other immunological disorders such as chronic viral infections like HIV and HTLV-I infection, Sjogren?s syndrome and thyroid disorders.
