The matrix metalloproteinases (MMPs) represent a family of more than 15 Zn-dependent ndopeptidases with extracellular activity. They share a common conserved protease domain sequence in which three His residues form a complex with a catalytic Zn ion.
MMP-13, also known as Collagenase-3 has a broad substrate specificity and plays potential role in tumor metastasis and invasion. The three-dimensional structure reveals a core domain for the protein consisting of three alphahelices and five beta-sheet strands with an overall tertiary fold similar to the catalytic domain of the other matrix metalloproteinase family members. MMP-13 has originally been identified in breast carcinomas. MMP-13 and the expression of the enzyme is upregulated under conditions that promote endothelial cell growth and vascular differentiation. The over-expression of MMP-13 associated with nonhealing wounds is shown for chronic dermal ulcers. There is growing body of evidence that MMP-13 is a major player in the disease condition of rheumatoid arthritis and osteoarthritis.
