The SH1 monoclonal antibody specifically binds to mouse Siglec-G (sialic acid binding immunoglobulin-like lectin G), also known as
Siglec10. Siglec-G is a Type I transmembrane glycoprotein that belongs to the Immunoglobulin superfamily. Siglec-G functions as an
adhesion molecule that mediates sialic-acid dependent binding to cells. Siglec-G is expressed on cells of the B cell lineage. Siglec-G is most
highly expressed by pre-B cells and B1a cells within the B cell lineage and is not detectable on T cells. Siglec-G can inhibit B cell
receptor-mediated calcium signaling when it is overexpressed. Mice that lack Siglec-G have significantly increased numbers of B1a cells that
begins early in development and is B cell intrinsic. Siglec-G-deficient mice have higher titers of natural IgM antibodies than their normal
counterparts. Mouse Siglec-G is the ortholog of human Siglec-10 (also known as, CD330).
This antibody is conjugated to BD Horizon™ PE-CF594, which has been developed exclusively by BD Biosciences as a better alternative to
PE-Texas Red®. PE-CF594 excites and emits at similar wavelengths to PE-Texas Red® yet exhibits improved brightness and spectral
characteristics. Due to PE having maximal absorption peaks at 496 nm and 564 nm, PE-CF594 can be excited by the blue (488-nm), green
(532-nm) and yellow-green (561-nm) lasers and can be detected with the same filter set as PE-Texas Red® (eg 610/20-nm filter).
