UCHT1单克隆抗体特异性结合人CD3ε链。CD3ε链是CD3/T细胞抗原受体复合物的一个20 kDa的亚基。70-80％的正常人外周血淋巴细胞和60-85％胸腺细胞都表达CD3ε。研究表明，与共刺激剂（例如美洲商陆有丝分裂原或抗CD28抗体）一起使用时，该抗体对CD3ε阳性细胞具有促有丝分裂作用。CD3在抗原识别过程的信号传导中起到核心作用。UCHT1抗体可同时将细胞表面和细胞内的CD3ε染色，另一个抗CD3抗体HIT3a，仅可将细胞外的CD3ε染色。

The UCHT1 monoclonal antibody specifically binds to the human CD3ε-chain, a 20-kDa subunit of the CD3/T cell antigen receptor complex. CD3ε is expressed on 70-80% of normal human peripheral blood lymphocytes and 60-85% of thymocytes. Studies from the HLDA Workshop show that this antibody is mitogenic for CD3ε-positive cells when used in conjunction with costimulatory agents such as pokeweed mitogen or anti-CD28 antibody. CD3 plays a central role in signal transduction during antigen recognition. The UCHT1 antibody stains both surface and intracellular CD3ε unlike the other CD3 clone, HIT3a, that stains only extracellular CD3ε.

1.This reagent has been pre-diluted for use at the recommended Volume per Test. We typically use 1 × 10^6 cells in a 100-μl experimental sample (a test).
2. An isotype control should be used at the same concentration as the antibody of interest.

3.Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.
4.The monoclonal antibody was purified from tissue culture supernatant or ascites by affinity chromatography.
5.The antibody was conjugated with PerCP-Cy5.5 under optimum conditions, and unconjugated antibody and free PerCP-Cy5.5 were removed. Storage of PerCP-Cy5.5 conjugates in unoptimized diluent is not recommended and may result in loss of signal intensity.
