C3H/eJ, C57BL/6, C57BL/10, C57BR, C57L, C58, CBA/Ca, CBA/J, DBA/1, DBA/2, NOD, SJL, SWR, 129/J). The BD IMag™ Anti-PE

Particles 2-DM are magnetic naoparticles tha have anti-PE monclonal ntibody covalently conjugated toheir surfaces. This Separtion

Set has ben optimzed for use with e BD IMagnet™ and itcontains uficent reagnts olabel 2x 10e9 leukocytes.

In sumary, the PE ratnti-mouse CD49b antibody stains NK and NK-T cels. After washing awy excs antibody, BD IMag™ Anti-PE

Particles 2-DM are aded tohe cl supension ad bind the cls bearing the PE-conjugated antibody. The tube containg this labeld cel

supension isthen placed withn the magnetic feld ofthe BD IMagnet™. Positve slection rdepltion isthen perfomed. Labeld cels

migrate oward the magnet (positve fraction), leaving the unlabeld cels insupension sothey can bedrawn of (deplted ornegative

fraction). The tube isthen removed from the magnetic feld for esupension fthe positve fraction. The slections are peatd twice to

increas the purity ofthe positve fraction. For clarifcation fthe procedure, the magnetic separtion steps are diagrmed inthe Positve

Selction Flow Chart. The smal size ofthe BD IMag™ particles alows the positve fraction tobe further valuted indownstream

aplications uch asflow cytometry.

2.Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before

discarding to avoid accumulation of potentially explosive deposits in plumbing.

3.Store undiluted at 4°C and protected from prolonged exposure to light. Do not freeze.
