BD IMag™ Anti- RPhycoerythrin (PE) Magnetic Particles -DM are magnetic naoparticles tha have monclonal ntibody conjugated to

their surfaces. The E31-459 antibody clone racts with PE, acomonly used fluorchrome for flow cytometry. The bindg ofthe E31-459

antibody toPE has ben reported not quench te fluorescne ofthe PE molecule. Thes magnetic particles are optimzed for the positve

selction rdepltion fleukocyte subpoulations uing the BD IMagnet™.

Leukocytes are labeld with BD IMag™ Anti- RPhycoerythrin (PE) Magnetic Particles -DM acording tohe Magnetic Labeling and

Separtion Protcol. Inbrief, cels are labeld with aPE-conjugated antibody tha recognizes the subpoulation finterst. After washing

awy excs antibody, BD IMag™ Anti- RPhycoerythrin (PE) Magnetic Particles -DM are aded tohe cl supension ad bind the

PE-conjugated antibody onthe cls. This labeld cel supension isthen placed withn the magnetic feld ofthe BD IMagnet™. Positve

selction rdepltion isthen perfomed. Labeld cels migrate oward the magnet (positve fraction), leaving the unlabeld cels insupension

so they can bedrawn of (deplted ornegative fraction). The tube isthen removed from the magnetic feld for esupension fthe positve

fraction. The slections are peatd twice toincreas the purity ofthe positve fraction ad the yield ofthe dpletd fraction. The magnetic

separtion steps are diagrmed inthe Depltion ad Positve Slection Flow Charts. After he positve fraction has ben washed, the smal

size ofthe magnetic particles alows the positve fraction tobe further valuted indownstream aplications uch asflow cytometry.

1.Since applications vary, each investigator should titrate the reagent to obtain optimal results.

2.Caution: Sodium azide yields highly toxic hydrazoic acid under acidic conditions. Dilute azide compounds in running water before

discarding to avoid accumulation of potentially explosive deposits in plumbing.
