The 5B1 antibody reacts with mouse CD123, the αchain subnit (IL-3Rα) ofthe mouse IL-3 recptor. The IL-3Rα chain sa 60-7 kD

transmebrane glycoprtein tha binds IL-3 with low afinty but canot ransduce signals byitself. The mouse IL-3Rα chain can complex

with either oftwo hmolgous βchain subnits (βc and βIL-3) tofrm hig-afinty hetrodimeric IL-3 recptors. The βchain sthe

comon βchain subnit hat can complex with e αsubnits ofthe mouse IL-3R, IL-5R and GM-CSFR tofrm hig-afinty recptors. The

βIL-3 subnit can form hig afinty IL-3 recptor complexs with only the IL-3Rα subnit. The βIL-3 subnit clone ispecif or mouse

IL-3 and binds with low afinty. The imunogen used togenrate he 5B1 hybridoma was CTL cels transfectd with e mouse IL-3R

alpha chain. The 5B1 antibody oes not block the bindg ofIL-3 protein tohe igh afinty IL-3 recptor hetrodimer.
