The Ambion® ToTALLY RNA™ Total RNA Isolation Kit is similar to the widely used guanidine thiocyanate, acid phenol:chloroform procedure of Chomczynski and Sacchi. All reagents needed to isolate, analyze, and store RNA (other than isopropanol) are included, making the kit an excellent value. The procedure has been modified to include an optional LiCl extraction step to reduce or eliminate DNA, carbohydrate, heme, and other contaminants that can be a problem with some guanidine-based methods. All reagents are RNase-free. The ToTALLY RNA™ Kit produces high-quality intact RNA from a wide range of sources, including plant tissues and bacteria. The purified RNA may be used for analysis by Northern blot, RNase protection, primer extension, RT-PCR, in vitro translation, and for construction of cDNA libraries. The kit contains reagents sufficient for the isolation of RNA from 10 g tissue or approximately 109 cells in culture.

Isolate RNA and DNA Sequentially From the Same Biological Sample
For many researchers, biological specimens are in short supply (e.g., tumor biopsy, scarce tissues) or are difficult and expensive to propagate (i.e., tissue culture cell lines). It is desirable in such cases to maximize their usefulness by extracting both the RNA and DNA fractions from the same sample. This can be done by adapting the protocol for RNA isolation used in Ambion's ToTALLY RNA™ Kit as outlined in Technical Bulletin #161, "Simultaneous Extraction of RNA and DNA" (see link above left).
