AHP987 recognises an epitope within the C-terminal (CT) region of glutamate receptor subunits 2 and 3 (GluR2/3), components of the AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid) group of ionotropic glutamate receptors, which play a key role at excitatory synapses.
The interaction of the C-terminal amino acids (-SVK1) of AMPA receptor GluR2/3 subunits with PDZ domain-containing proteins, has been implicated in the regulation of synaptic targeting and surface expression of AMPA receptors. Three such PDZ domain-containing proteins identified, are glutamate receptor interacting protein 1 (GRIP1), AMPA receptor-binding protein (ABP/GRIP2) and protein interacting with C-kinase 1 (PICK1).
Studies into cerebellar long-term depression (LTD) have indicated the requirement for PKC-regulated interactions between the C-terminal of GluR2/3 and PDZ domain-containing proteins(1,3).
