AHP1917 specifically recognises human Osteoprotegerin (OPG), an osteoblast- secreted protein and member of the TNF Receptor superfamily (TNFRSF11B), which neutralizes the function of sRANKL/CD254 in osteoclastogenesis, by acting as a decoy receptor, blocking the signal between stromal cells and osteoclastic progenitor cells.
OPG is highly expressed in a wide range of tissues, including adult heart, lung, liver, prostate, ovary and bone marrow. The ligand for OPG, sRANKL/CD254, also known as OPGL (TNFSF11) or ODF (Osteoclast differentiation factor), acts as a potent inducer of bone resorptionin vivo, and in conjuction with M-CSF, stimulates osteoclast development and activates mature pre-existing osteoclasts.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.

Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Antiserum Preparation
Antiserum to human OPG were raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.
