AHP1888T recognises mouse Dact1 (Dapper homolog 1), an intracellular protein and member of the dapper family, which acts as a positive regulator of the signalling pathways of Dishevelled 2 (DVL2), a cytoplasmic protein which plays a pivotal role in Wnt signalling during vertebrate development.
Dact1 is involved in enhancement of the transcriptional activation of target genes of the Wnt signalling pathway, through its intracellular co-localization with DVL2 and inhibition of the degradation of CTNNB1/beta-catenin. Studies have shown that mice homozygous for mutations in Dact1, display malformations involving the spine, genitourinary system and distal digestive tract, which was traced to disrupted germ-layer morphogenesis at the primitive streak, seeSuriben, R.et al.for details.
