AHP1721 recognises the C-terminus of phosphoinositide 3-kinase enhancer (PIKE). It exists as isoform 1 (PIKE-L) and isoform 2 (PIKE-A). PIKE-L is the longer of the two (124kDa) and participates in the prevention of neuronal apoptosis by enhancing PI3 kinase activity. It does so by binding to the adaptor protein Homer, linking to metabotropic glutamate receptors. The shorter PIKE-A isoform is also known as Centaurin gamma 1. It does not stimulate PI3 kinase but may protect cells from apoptosis by stimulating Akt. Overexpression of PIKE-A enhances Akt activity and promotes cancer cell invasion. PIKE-A also regulates the adaptor protein 1-dependent trafficking of proteins in the endosomal system. PIKE-L is found in the cytoplasm and nucleus, and is brain-specific, whereas PIKE-A is ubiquitously expressed and found in the cytoplasm only.
AHP1721 recognises both PIKE-L and PIKE-A.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.Sodium citrate buffer pH 6.0 is recommended for this purpose.

Antiserum to PIKE (CT) was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by ion exchange chromatography.
