AHP1695 recognises Human Damage-regulated autophagy modulator (DRAM).
DRAM is a p53 target gene encoding a lysosomal protein that induces autophagy, a process that degrades cytosolic proteins and organelles.
Research indicates that the activation of DRAM by p53 is simultaneous to the activation of p53 of one or more pro-apoptotic genes (e.g PUMA, Bax ), and that the signalling pathways regulated by these genes together promote a full cell death response.
DRAM, alone, is unable to induce apoptosis, however research has shown that it is inactivated in certain cancers. This highlights the importance of DRAM and suggests that autophagy may play a more important role in cancer than originally believed.
It has been demonstrated that DRAM exists in a number of isoforms.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.Sodium citrate buffer pH 6.0 is recommended for this purpose.
Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.
Western Blotting
AHP1695 detects a band of approximately 31kDa in K562 cell lysate.
Storage
Store at +4oC or at -20oC if preferred.
Storage in frost-free freezers is not recommended.
This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life
18 months from date of despatch.
Antiserum Preparation
Antisera to Human DRAM were raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG prepared from whole serum by affinity chromatography .
