AHP1521 recognises src homology 2 domain containing tyrosine phosphatase (SHP2), also known as PTPN11, a 68 kDa member of the protein tyrosine phosphatase family.
SHP2 acts downstream of growth factor receptors and cytoplasmic protein tyrosine kinases which transduce signals from the cell surface to the nucleus. The recruitment and activation of SHP2 is mediated through the binding of CD172a to its ligand CD47. SHP2 regulates a variety of cellular processes. These include transcription, embryonic development, cell growth, cell migration, differentiation, mitotic cycle, haematopoiesis and oncogenic transformation. As a result, dysregulation of the SHP2 gene has been linked to LEOPARD, Noonan and Noonan-like syndrome as well as acute myeloid leukaemia. SHP2 is ubiquitously expressed, with highest levels found in heart, brain, and skeletal muscle.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.Sodium citrate buffer pH 6.0 is recommended for this purpose.

This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Antiserum to human SHP2 was raised by repeated immunisation of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by affinity chromatography.
