AHP1474 specifically recognises human CoREST (REST corepressor 1), a ubiquitously expressed member of the CoREST family and component of the BHC (BRAF–HDAC) transcriptional corepressor complex, involved in the repression of neuronal-specific gene transcription.
The regulation of neuronal cell specific gene expression, is dependent on the repression of non-neuronal cell gene transcription, mediated by the neuronal silencer protein REST/NRSF (RE1 silencing transcription factor/neural-restrictive silencing factor) through recruitment of the BHC complex. CoREST is a SANT-domain-containing protein within the BHC complex, which mediates the nucleosomal demethylation of H3K4 (histone H3 lysine 4), and acts as a regulator of BHC corepressor function.
