AHP1444 recognises Lissencephaly 1 (LIS1), also known as Platelet-Activating Factor Acetylhydrolase Isoform 1B Alpha Subunit or PAFAH, a 47kDa enzyme essential for precise control of mitotic spindle orientation in neuroepithelial stem cells and radial glial progenitor cells. The protein is highly conserved, with near-identity between mouse and humans. It is expressed predominantly in the brain and regulates tubulin to suppress microtubule dynamics. It participates in cytoplasmic dynein-mediated nucleokinesis, somal translocation and cell motility as well as mitosis (neurogenesis) and chromosome segregation.
Lissencephaly or ‘smooth brain’ is a brain malformation characterised by a smooth or nearly smooth cerebral surface. It has many causes, but mutations in the LIS1 gene, triggering disruption in the migration of post-mitotic neurons during gestation, causes classical lissencephaly. Symptoms might include unusual facial appearance, psychomotor retardation and seizures.
