AHP1347 detects E2F1 phosphorylated at serine 364 (pSer364). E2F1 is a transcription activator that binds DNA cooperatively with the differentiation regulated transcription factor proteins (DP). Together they bind to the promoter region of a number of genes whose products are involved in cell cycle regulation or DNA replication. E2F1 also binds to the retinoblastoma protein (RB1), which functions in the control of cell-cycle progression from G1 to S phase.
E2F1 can also mediate p53-dependent and independent apoptosis, and phosphorylation of E2F1 at serine 364 results in its stabilisation and apoptosis induction.
E2F1 is a target of the transforming proteins of small DNA tumour viruses and increased expression of this protein has been reported in a variety of cancers.

For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
