AHP1307 specifically recognises the 55kDa Tryptophan Hydroxylase 2 (TPH2) protein, when phosphorylated at Ser19.
Tryptophan Hydroxylase (TPH) is a rate-limiting enzyme which catalyses the 5-hydroxylation of tryptophan, to form the amino acid 5-hydroxytryptophan (5-HTP), in the first stage of the biosynthesis of the neurotransmitter serotonin (5-hydroxytryptamine/5HT). Mammalian TPH is encoded by two genes: TPH1 expressed in peripheral tissues e.g. skin and gut, and TPH2 primarily expressed in the brain. Studies have demonstrated the regulation/enhancement of TPH activity through phosphorylation by cAMP-dependent protein kinase (PKA) and Ca2+/calmodulin kinase II (CaMKII), the latter of which phosphorylates a Ser19 site within the regulatory domain of TPH2.

For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
