AHP1304 specifically recognises the 52kDa parkin protein, when phosphorylated at Ser101.
Parkin is an E3 ubiquitin ligase highly expressed in the brain, which functions in the ubiquitin-proteasome system, together with the E2 ubiquitin conjugating co-enzyme UbcH7. Mutations in the parkin gene are associated with the hereditary neurodegenerative disorder known as autosomal recessive early onset Parkinson disease 2 (PARK2)/autosomal recessive juvenile Parkinson disease (PDJ), characterized by progressive dopaminergic neural cell death. Studies have identified the presence of at least five phosphorylation sites in parkin including Ser101, shown to be phosphorylated by Casein kinase-1 (CK-1), and suggest that phosphorylation of parkin may act to regulate its ubiquitin ligase activity.

For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
