AHP1299 specifically recognises the 29kDa integral membrane protein Aquaporin 2 (AQP2), when phosphorylated at Ser261.
AQP2 provides the plasma membranes of the renal collecting duct with a high permeability channel to enable the movement of water across an osmotic gradient, under hormonal regulation by vasopressin. In addition to the Ser256 phosphorylation site, studies have identified a novel phosphorylated Ser261 site on AQP2, which decreases in expression, in response to vasopressin.
Gene mutations in AQP2 result in hereditary nephrogenic diabetes insipidus in humans, and acquired diabetes insipidus can also occur through a decrease in the expression of the AQP2 gene, following lithium treatment. Expression of the AQP2 gene is increased during pregnancy and other conditions associated with water retention.

For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.
Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. AHP1299 detects bands of approximately 29kDa and 37kDa (glycosylated form) in rat kidney cell lysates.

Storage in frost-free freezers is not recommended.
This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Antisera to rat aquaporin 2 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
