AHP1287 specifically recognises Wnt-1, formerly known as int-1, a 343 amino acid non-glycosylated secreted protein associated with the extracellular matrix, involved in normal embryonic development and particularly linked with breast cancer.
Wnt-1, which signals through binding to the Frizzled family of transmembrane receptors, was originally identified as a proto-oncogene implicated in carcinomas induced by the mouse mammary tumour virus (MMTV). Wnt-1 belongs to a family of Wnt genes involved in the dorso-ventricular development of the mesoderm during embryogenesis, with a restricted temporal and spatial pattern of expression.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.
Storage
Prior to reconstitution store at +4oC.
After reconstitution store at -20oC.
Storage in frost-free freezers is not recommended. This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life
12 months from date of reconstitution.
Antiserum Preparation
Antisera to human Wnt-1 were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared by affinity chromatography.
