AHP1253 recognises amyloid precursor protein (APP), a 130 kDa member of the APP family, expressed in adult brain. It is a cell surface kinesin I membrane receptor which regulates the axonal transport of beta-secretase and presenilin 1.In Alzheimer's disease processing of APP by beta-secretase gives rise to amyloidogenic 42 AA beta amyloid protein that is found in senile plaques and vessels. APP binds components of the extracellular matrix such as heparin and collagen I and IV, regulating neurite growth and is also involved in neuronal adhesion, cell mobility and axonogenesis. Through it’s binding to APBB1/Tip60, APP plays a role in the regulation of transcription. APP also inhibits Notch signaling through it’s interaction with NUMB.
