AHP1242 detects Apoptosis Signal-regulating Kinase 1 (ASK1) phosphorylated at serine83. ASK1 is a serine/threonine kinase also known as MAP kinase kinase kinase (MAPKKK), and plays an important role in the apoptotic response to various stimuli including oxidative stress, cytokines (e.g. TNF) and endoplasmic reticulum stress. Akt in conjunction to Hsp90 phosphorylates ASK1 at Ser83 to inhibit ASK1 induced apoptosis.

For the detection of phosphoproteins, tyrosine phosphatase inhibitors such as 1mM Sodium Orthovanadate should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background.

This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.

Antisera to ASK1 (pSER83) were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG was prepared from whole serum by ion exchange chromatography.
