AHP1189 recognises 37kDa polycomb complex protein BMI-1, involved in segment specification, cell growth and maintenance, transcriptional regulation and chromatin modification. BMI-1 is a proto-oncogene which cooperates with the myc oncogene to produce B-lymphomas. It is required in the regulation of normal adult and leukaemic stem cells, making BMI-1 a potential target for stem cell proliferation and anti-cancer therapies. BMI-1 represses transcription at the ink4 locus which contains tumour suppressors p16 and p19Arf.
BMI-1 is a component of the Polycomb group (PcG) multiprotein PRC1 complex which ubiquitinates histone-2A at lys119, resulting in a heritable change in the ability of chromatin to express genes. BMI-1 is also required to stimulate the E3 ubiquitin ligase activity of RNF2/RING2 within the PRC1 complex.
Recent studies have shown that the Notch ligand DLL4 (delta-like protein 4) enhances T cell proliferation, by directly influencing BMI-1 transcription.
