AHP1015 recognises NOD2, an intracellular protein of 100kDa, which belongs to the NOD (nucleotide binding oligomerisation domain) family. The NOD family comprises at present more than 20 different mammalian NOD-LRR (leucine-rich repeat) proteins, which are involved in the regulation of apoptosis and the immune response.
NOD2 has been shown to be constitutively or inducibly expressed in monocytes, macrophages, T and B cells, dendritic cells, as well as intestinal epithelial cells. A specific motif of peptidoglycan (PGN), muramyl dipeptide (MDP), has been identified as the sole ligand of NOD2.
NOD2 and Toll-like receptors (TLR) are emerging as key mediators of innate host defence in the intestinal mucosa, crucially involved in maintaining mucosal and commensal homeostasis.
Mutations in the gene encoding NOD2 are a major susceptibility factor underlying a significant subgroup of patients with Crohn’s disease.
