AHP1011 recognises human clusterin (also known as apolipoprotein J). Clusterin is a ubiquitous glycoprotein which exists in at least two distinct isoforms; a proapoptotic nuclear form, of 49kDa, and a prosurvival secretory form with a molecular weight of 75-80kDa. Clusterin is implicated in several diverse physiological processes including sperm maturation, cell differentiation, and apoptosis.
Clusterin upregulation has been reported in various human malignancies, including bladder, prostate, and breast tumours.
Following specific stress induced injury, secretory clusterin acts as a molecular chaperone, scavenging denatured proteins outside cells. It has been shown to be upregulated in Alzheimer’s disease, retinitis pigmentosa, glomerulonephritis, atherosclerosis and myocardial infarction.

This product requires antigen retrieval using heat treatment prior to staining of paraffin sections. Sodium citrate buffer pH 6.0 is recommended for this purpose.
Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using appropriate negative/positive controls.
Western Blotting
AHP1011 detects a band of approximately 60kDa in human brain tissue lysate.
Storage
Store at +4oC or at -20oC if preferred.
Storage in frost-free freezers is not recommended.
This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
Shelf Life
18 months from date of despatch.
Antiserum Preparation
Antisera to Clusterin were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG prepared by affinity chromatography.
