Background/Target Information
JAK 3 (also designated L JAK) is a member of the Janus family of protein tyrosine kinases along with JAK 1, JAK 2, or Tyk 2. Subsequent to activation, these kinases function to activate various members of the Stat family of transcription factors by phosphorylation on critical tyrosine regulatory sites. JAK 3 is activated in response to IL-3 and IL-4 in T cells and myeloid cells. JAK 3 activation requires the serine rich membrane proximal region of the IL 2 receptor beta chain, but does not require the acidic domain that is required for association and activation of Src family kinases. Expression of JAK 3 is restricted to NK and T cells.
