Invitrogen™ Phasemaker™ Tubes contain a thick liquid polymer that separates and completely isolates the upper aqueous phase of a TRIzol® Reagent mix from the organic phase underneath, greatly simplifying removal of the upper phase containing the RNA.

The polymer in Phasemaker Tubes is heavier than the aqueous phase of the TRIzol Reagent mix, but lighter than the organic phase, so after centrifugation it positions itself between these two layers. As a thick polymer, it creates a tight seal between the two phases, easing removal of the aqueous phase and helping to reduce the risk of carrying over contaminants from the organic layer, thereby increasing RNA recovery up to 30%.
The polymer in the Phasemaker Tubes is inert and does not interfere with standard RNA applications. Isolated RNA can be used in RT-PCR, Northern blot analysis, dot blot hybridization, poly(A)+ selection, in vitro translation, RNase protection assay, and molecular cloning.
Phasemaker Tubes accommodate RNA extraction using TRIzol Reagent, TRIzol LS Reagent, and TRIzol Plus Purification Kit only. They are not recommended for use with other organic reagents as the density of those reagents might not be suitable. Phasemaker Tubes are not for use in DNA or protein isolation, but are intended for RNA extraction only.
Phasemaker Tubes work with fresh or frozen samples, including tissue and blood. They also work with samples previously treated with RNAlater™ RNA stabilization solution. Phasemaker Tubes are already pre-dispensed; the tube size is 2 mL.
