4670-5107 recognises the glutamate receptor 1 (also known as AMPA) which acts as an excitatory neurotransmitter at many synapses in the central nervous system. Binding of the excitatory neurotransmitter L-glutamate induces a conformation change, leading to the opening of the cation channel, and thereby converts the chemical signal to an electrical impulse. The receptor then desensitizes rapidly and enters a transient inactive state, characterized by the presence of bound agonist.
This antibody linked to immobilized Protein A immunoprecipitates detergent-solubilized receptor from brain or transfected cells.

Where this product has not been tested for use in a particular technique this does not necessarily exclude its use in such procedures. Suggested working dilutions are given as a guide only. It is recommended that the user titrates the product for use in their own system using the appropriate negative/positive controls.
Immunohistology
Paraformaldehyde or paraformaldehyde/glutaraldehyde are suitable. Cryostat and vibratome sections can be used with or without Triton X-100 treatment.
Western Blotting
Western blot analysis of rat brain shows a single band of approximately 110 kDa co-migrating with GluR1 expressed in transfected cells.
Storage
Store at +4oC or at -20oC if preferred.
Storage in frost-free freezers is not recommended.
This product should be stored undiluted. Avoid repeated freezing and thawing as this may denature the antibody. Should this product contain a precipitate we recommend microcentrifugation before use.
