This antibody reacts with an internal region of the ZONAB protein. On Western blots, it identifies a major band at ~65 kDa and a minor band at ~54 kDa. The antibody is directed towards a region of ZONAB common to both the long and the alternatively-spliced short form. During development, reactivity was confirmed using mouse testis tissue homogenates by Western blotting and labeling of mouse testis tissue using immunofluorescence. All signals were absent in knockout mice, further confirming the reactivity with ZONAB.
Background/Target Information
ZONAB (ZO-1 associated nucleic acid-binding) is a Y-box transcription factor whose nuclear localization and transcriptional activity is regulated by the tight junction-associated candidate tumor suppressor ZO-1. Two ZONAB isoforms are known, a long isoform (ZONAB B) and an alternatively spliced shorter isoform (ZONAB A) that lacks 68 amino acids. ZONAB regulates the expression of the tyrosine kinase receptor HER2, and appears to act in combination with ZO-1 to regulate HER2 expression.
