2115-5004 is specific for clathrin and stains coated vesicles from various organs and tissues tested e.g. brain, mammary gland, ovaries. It also reacts with the cell lines, HeLa, SV-40 and RVF-SMC.
Clathrin is a protein which assembles into a polyhedral network on the cell membrane as the membrane invaginates, forming a coated pit which is essential to endocytosis. It is composed of three polypeptides, a 180 kDa heavy chain and two 32-38 kDa light chains which combine to create a distinct three-legged triskelion. It is this morphology which allows clathrin to form its unique polyhedral network.
In nervous tissue, 2115-5004 cross-reacts with another coated vesicle protein of 190 kDa which is not present in isolated clathrin triskelions. In some epithelial cells of certain species, this antibody may react with a cytokeratin component.
