The CD28 antibody recognizes the human CD28 antigen, which is highly expressed on CD3+ thymocytes, most peripheral thymocytes and plasma cells. Binding of CD28 to its ligands CD80 or CD86 costimulates T cell effector function and T cell–dependent antibody production in vitro and in vivo.
In the presence of antibodies directed against CD2 and CD3, CD28 antibodies stimulate T cell proliferation and cytokine production.
Activation and proliferation of T cells can be induced by interaction of the T cell receptor with peptide-MHC complexes expressed on the surface of APCs. The T cell receives a signal transduced through the CD3 complex. Cytokines or other costimulatory signals are required in addition. Activated T cells can be used for any downstream processes, such as cytokine analysis or immunoprecipitation. Cells can also be transfected with high efficiency.
